Video 1
Ionomycin-induced
translocation of myrVILIP-1-GFP in COS-7 cells and complete reversal
after calcium depletion with EGTA. Living COS-7 cells expressing myrVILIP-1-GFP
were stimulated by addition of 2 µM ionomycin (+IONOMYCIN) leading to
increased fluorescence at the plasma membrane and intracellular structures. The
ionomycin-dependent localization of myrVILIP-1-GFP was reversed
after addition of 5 mM EGTA (+EGTA) to lower the calcium concentration in the
incubation buffer. Fluorescence of myrVILIP-1-GFP-transfected COS-7
cells was monitored by taking images every 30 seconds using a Leica DMR upright
fluorescence microscope and a Visitron CCD camera.
