Reversible translocation of myrVILIP-1-GFP to the plasma membrane and subcellular compartments in primary hippocampal neurons after addition of 50 mM KCl to the stimulation buffer (+ KCl) leading to the concentration of green fluorescence at specific sites of the plasma membrane and of intracellular membranes. Addition of 5 mM EGTA (+ EGTA) leading to depletion of calcium from the stimulation buffer shows the complete reversibility of myrVILIP-1-GFP translocation. Fluorescence of myrVILIP-1-GFP-transfected hippocampal neurons was monitored by taking images every 10 seconds using a Leica DMR upright fluorescence microscope and a Visitron CCD camera.
