A, B, Efficient gene transfer with adenoviral vectors was confirmed by Ax-LacZ in a Schwann cell monoculture (A) compared with the control group (B). C, D, In coculture with DRG neurons, Ax-GFP was used to observe efficiency of gene transfer to Schwann cells. Note that none of the neurons in the phase contrast image (D, arrow) showed green fluorescence. E, F, -Galactosidase (-gal) staining of Ax-LacZ-infected nerve graft before implantation.   • Supplemental Fig.2 - Supplemental Figure 2. Myelination was equivalent at the distal part to the nerve grafts. Five weeks after transplantation, specimens were taken from the proximal part of the grafts (A, B) and from the host nerves distal to the grafts (C, D). A and C were taken from the same Ax-LacZ-treated recipient rat, and B and D were taken from an Ax-myrAkt-treated rat. Immunohistochemistry with anti-MBP antibody (A--D) shows the equivalent extent of myelination at a point distal to the grafts (C, D), whereas the proximal section (A, B) shows increased myelination in the Ax-myrAkt-treated rat. Scale bar, 200 m.
  • Supplemental Fig.3 - Supplemental Figure 3. BrdU-labeled implanted cells form a myelin sheath. Transplantation was performed with a BrdU-labeled nerve graft and fixed 2 weeks later. Arrowheads indicate BrdU-positive (green) implanted cells, which are adjacent to the MBP-positive (red) myelin sheath. At this time point, myelin formation was immature, and a clear ring shape of myelin was not always observed. Scale bar, 5 m.
