  • supplemental material - Supplemental Figure 1. PDGFR? Expression by Retinal Astrocytes Immunohistochemical analyses showing the expression pattern of PDGFR? (A, C) in relation to that of Pax2 (B, C). Pax2 is specific for cells in the astrocyte but not oligodendrocyte lineage, microglial cells, endothelial cells, or meningeal cells in E17.5 wild-type retinas. These results show that PDGFR? can be used reliably to define astrocytes in the developing retina. Scale bar; 80?m.

Retinal Cell Subtype Identities in Tlx KO Retinas Immunohistochemical analyses of wild-type (A, C, E, G, I, K, M, O) and Tlx KO (B, D, F, H, J, L, N, P) retinas at 1 month after birth, using antibodies against markers that reliably detect each of the retinal cell types: (A, B) Brn3b; (C, D) HPC-1; (E, F) PKC; (G, H) calbindin; (I, J) Ret P-1;(K, L) PNA; (M, N) GS; (O, P) GFAP (see text for details). (A-L) Each retinal neuronal subtype was detected with its specific antibody, however, the numbers of each cell type appeared to be reduced in Tlx KO retinas compared to wild-type. (M-P) In Tlx KO retinas, GS, a specific marker of Müller cells, was detected in a similar pattern to wild-type. On the other hand, GFAP, an astrocyte marker that is localized to the inner retinal surface in wild type (arrows in O), was detected in a pattern characteristic to Müller cells (arrowheads in P) in Tlx KO retinas, suggesting that Müller cells underwent reactive GFAP expression. Cell nuclei were stained with TO-PRO-3 iodide (blue). Scale bars; 80 ?m.
