Fluorescent Proteins Expressed in Mouse Transgenic Lines Mark Subsets of Glia, Neurons, Macrophages, and Dendritic Cells for Vital Examination
J. Neurosci. Zuo et al. 24: 10999

Supplemental data

Files in this Data Supplement:

  • supplemental material - Figure S1. YFP-expressing cells in the muscle and nerve are macrophages. In a whole mount of the triangularis sterni muscle from a YH animal, a YFP+ cell (A) is labeled by the antibody F4/80 (B). C, D, In the distal stump of the plantar nerve of a YH animal, YFP-expressing cells are present 14 days after resection of the sciatic nerve. These cells have a rounded, phagocytic morphology (C) and are labeled with an antibody to the macrophage marker FA-11 (D). Bar, 30 |*mu*|m.
  • supplemental material - Figure S2. YFP labels dendritic cells in the gut. Confocal images of cells in a cryptopatch at the base of a villus of the small intestine of a YF animal. Some YFP-expressing cells (arrows in A) are labeled by an antibody to a marker for dendritic cells (anti-CD11c, arrows in B), while other YFP+ cells are negative for CD11c (arrowhead), and some CD11c+ cells are YFP- (double arrowhead). Together with similar findings (not shown) for YFP+ cells probed with a macrophage marker, these results show that subsets of dendritic cells and macrophages are labeled with YFP. Bar, 10 |*mu*|m.
  • supplemental material - Figure S3. In the Kosmos line, GFP marks astrocytes. A, Adult hippocampus. B, Adult cerebellum. In these coronal sections, brightly fluorescent cell bodies of astrocytes punctuate a background of dense astrocytic processes and unstained areas that are the cell bodies of neurons. The most prominent labeling in the cerebellum is in the Bergmann glia. Bar, 250 |*mu*|m in A, 1 mm in B.
  • supplemental material - Figure S4. Some transgenic lines show distinct patterns of astrocyte and microglial labeling. Confocal (A-C, G) and non-confocal (D-F) images of sections of adult brain. A-C, Cortex of Kosmos line. An astrocyte labeled with YFP (A) is also positive for GFAP (B); merged image of A and B with the GFAP label pseudocolored red and YFP green (C). D-F, Microglial cell labeled with YFP (D) is also labeled with anti-F4/80 (E). F, Merged image of C and D with the F4/80 label pseudocolored red and YFP green. G, YFP labels a mixture of astrocytes and microglia in the cortex of the YH line. Microglia are intensely labeled and astrocytes more dimly labeled. Bar, 20 |*mu*|m in A-C, G. 35 |*mu*|m in D-F.
