Supplemental Figure 1. Efficient Recombination by CaMKII-cre Does Not Affect Spine or Synapse Number. Immunostaining for YFP in Rosa26-YFP x CaMKII-cre (a) and Rosa26-YFP x CaMKII-cre x TrkB flx/flx (b) mice reveals the same degree of recombination in animals having only one flox-flanked allele versus animals with three. Histograms and statistical analysis of spine density reveal no change between wild-type and TrkB flx/flx x CaMKII-cre animals (c; n=80 neurons from 2 animals and 60 neurons from one animal, respectively). TrkB x CaMKII-cre mice that are aged for one year do not develop a decrease in PSD-95 puncta density (d; 752.8 ± 74.9 puncta/mm2 versus 688.6 ± 61.2 puncta/mm2 from 2 control and 3 TrkB x CaMII-cre knockout mice respectively, p>0.55). No decrease in the number of synaptophysin positive puncta was detectable at one year of age, as well (e; 594.8 ± 103.5 puncta/mm2 versus 557.3 ± 12.6 puncta/mm2 from 2 control and 3 TrkB x CaMII-cre knockout mice respectively, p>0.68).
