SUPPLEMENTAL FIGURE 1: Schematic diagram depicting the behavior of PrP-EGFP and its interaction with PrPSc in scrapie-infected mice. (A) In wild-type (Prn-p+/+) mice, endogenous PrPC is converted to PrPSc. (B) In Tg(PrP-EGFP+/+)/Prn-p0/0 mice, PrP-EGFP cannot be converted to PrPSc-EGFP. (C) In Tg(PrP-EGFP+/0)/Prn-p+/+mice, PrP-EGFP binds to PrPSc formed from endogenous PrPC, and competes with endogenous PrPC for access to PrPSc, thereby slowing the conversion process. Because it is incorporated into growing deposits of PrPSc, PrP-EGFP marks the microscopic location of PrPSc in the brain.
