Supplemental Figure 1. Co-immunoprecipitation of cytosolic sortilin (cytSort) with BDNF. HEK 293 cells were co-transfected with cDNA’s encoding a mutant cytoplasmic region sortilin fused with GFP, as described in Materials and Methods, along with carboxy terminal tagged BDNF-HA. Cell lysates were immunoprecipitated with HA antibodies and immunoblotted with GFP antibodies (top panel). Immunoprecipitation of HA-BDNF was confirmed by immunoblotting with HA antibodies (middle panel). Crude lysates were immunoblotted with GFP antibodies to confirm equivalent expression levels (bottom panel).

Supplemental Figure 2. pH dependence of BDNF-sortilin interaction. HEK 293 cells were co-transfected with cDNA’s encoding sortilin and carboxy terminal HA-BDNFVal. Cell lysates were adjusted to the indicated pH’s, and immunoprecipitated with HA antibodies and immunoblotted with sortilin antibodies (top panel). Immunoprecipitation of BDNF-HA was confirmed by immunoblotting with HA antibodies (middle panel).
