  • supplemental material - Supplementary Figure 1. Selective loss of VAMP2 from TNTCFP­-expressing synapses. Dissociated hippocampal cultures were transfected with SYNYFP and TNTCFP before plating, cultured for 21 days, then fixed and stained for VAMP2 and synapsin. Active synapses formed by nontransfected axons show colocalization of synapsin and VAMP2 (arrowheads). Inhibited synapses, expressing SYNYFP and TNTCFP, lack staining for VAMP2, whereas staining for synapsin was unchanged (arrows). Scale bar=10 µm. We found punctate VAMP2 immunofluorescence present at 42% of nontransfected synapses defined by synapsin, but present at only 8% of TNTCFP-transfected synapses (n>150 synapses each condition, t-test p<0.01).
