Mitochondrial Nitric Oxide Mediates Decreased Vulnerability of Hippocampal Neurons from Immature Animals to NMDA

  • supplemental material - Supplementary Material. Time-lapse movie of NO production and localization in mitochondria of P5 neurons in response to NMDA (300 µM). Three neurons are in the field of view. Mitochondria were labeled with Mitofluor red 589, a ??m-independent mitochondrial marker (red). NO production was visualized with DAF-FM (green). Images were obtained sequentially every 20 seconds and digitally deconvolved to remove out-of-focus light. To visualize only the DAF-FM fluorescence produced during NMDA, the DAF-FM image obtained immediately prior to NMDA was subtracted from the subsequent images, and the subtracted images overlaid on the corresponding Mitofluor red 589 image. NMDA is perfused after the first image. Yellow pixels represent NO within mitochondria.
