Supplemental Figure 1. C280G mutant is localized to noncompact myelin in Gjb1-null mice. These are images of unfixed teased fibers from wild type (A-C) and TG + Gjb1-null mice line 7M33 (C280G, D-F). The fibers were immunostained with a rabbit antiserum against claudin-19 (red) and a mouse monoclonal antibody against Cx32 (green). Claudin-19 is localized similarly in both wild type and TG + fibers – to incisures (arrowheads), paranodes (the regions that flank the nodes of Ranvier – apposed arrowheads) as well as inner and outer mesaxons (arrows). In TG + fibers, Cx32 is localized to incisures and the outer mesaxons, in addition to the predominately paranodal localization in wild type fibers. Scale bars: 10 µM.

These are images of semi-thin sections of the sciatic nerve, (including a presumed motor fascicle) from TG + or TG - Gjb1 -/Y littermates (line 7M33 at P158). The myelinated axons appear normal in the TG + nerves, whereas the TG – nerve contains demyelinated axons (asterisk) and remyelinated (r), and macrophages containing myelin debris (m). The areas outlined by the rectangles in A and B are enlarged in C and D (but D is the mirror image of the rectangle region in B). Scale bars: 10 µM.
