The telencephalic hemispheres were placed between a platinumplate electrode (positive pole) and a tungsten needle electrode (negative pole) in PBS. Plasmid solution was injected into the MGE or CGE with a glass micropipette, and electronic pulses were generated.

(A, B) Coronal slices were prepared at 40 hours in vitro after focal electroporation into the MGE of E13.5 telencephalic hemispheres. The asterisk shows the place of electroporation. A significant number of the CGE-derived cells exhibited a clear migratory stream from the site of electroporation in the CGE to the hippocampus (See Figure 3 for details). The morphological features and migratory behavior of migrating CGE-derived cells resembled those of 'branching cells' (Nadarajah et al. 2003) in that they were highly motile in the formation and retraction of their processes.
