  • supplemental material - Hypothetical model for the role of REN in cerebellar GCP development. The developmentally-regulated increasing expression of REN (in association with p27) (blue and green co-expressing cells) along the transition from outer to inner EGL GCPs suppresses Shh signaling, thus lowering the intracellular levels of Gli1 (red) highly expressed in outer EGL. By this way, REN activity results in promoting growth arrest of inner EGL GCPs, allowing granule cells to complete the differentiation and migration processes into IGL. REN-induced apoptosis also contributes to limit the number of developing granule cells. Impairment of REN expression, as a consequence of 17p deletion, withdraws such a limiting signal for immature cell expansion, thus favoring tumorigenic events.
