A) Typical time courses (upper panel) of mGluRs or GABAb receptors mediated inhibition of EPSCs from excitatory MNTB synapses of P6 mice. The duration of each agonist application are indicated by horizontal bars near the top of the graph. Representative current traces of EPSCs before and after agonist application are shown in the lower panel. L-AP4 was applied alone (100 ?M) or in combination with ACPD (100 ?M) while baclofen was given at a concentration of 50 ?M in all experiments. B) At P18, there was no discernable effect of mGluR agonists on the EPSC; however agonist for GABAb still resulted in robust inhibition of EPSC amplitude, relative to control. Reduction in presynaptic inhibition is age-dependent. A summary of average presynaptic inhibition induced by L-AP4, LAP4 plus ACPD and baclofen from three age groups: P6-7, P9-10, and >P12. The extent of inhibition at each synapse was estimated by comparing the average amplitude of three to five consecutive EPSCs in the absence and presence of agonist(s). In each group, results from 6 to 12 recordings were pooled and compared (*p < 0.05 and ** P < 0.01, compared to P6-7).
