Figure 1. Hippocampi were rapidly removed from p4 rat pups and snap frozen in isopentane cooled in liquid nitrogen and stored at -80oC until processing. Cross sections were cut at 15um and fixed by immersion in 2% ice-cold paraformaldehyde for 30 seconds, followed by a wash in PBS and then immersion in freezing acetone (-18oC) for 30 seconds. Sections were washed in PBS for a further 5 minutes and were kept at -80oC before immunostaining as described for cultures (see Methods). Images obtained on a Zeiss 510 CLSM taken in areas dominated by glial cells. Blanks lacking either primary or secondary antibodies showed no significant signals under these conditions. The co-expression of both subunits of the NADPH oxidase with GFAP is evident.
