Hippocampal Neurons Express a Calcineurin-Activated Adenylyl Cyclase

  • supplemental material - Supplemental Figure 1. Calcium and forskolin-stimulated adenylyl cyclase activities in hippocampal membranes from calcineurin knockout and wild type mice are similar. Hippocampi were dissected from young adult wild type (Wt) or calcineurin knockout (CN-/-) mice. Membrane preparations were isolated and assayed for adenylyl cyclase activity as described in Materials and Methods in the presence of (A) various doses of free calcium (Ca2+), or (B) 50 µM forskolin (Forskolin). Error bars are standard deviations from triplicate determinations.
  • supplemental material - Supplemental Figure 2. Depolarization-induced cAMP accumulation in cultured wild type hippocampal neurons is dependent on phosphatase activities. Cultured hippocampal neurons from wild type newborn pups were maintained for 14 days before assayed for cAMP accumulation as described in the Materials and Methods. Neurons were pretreated with the carrier for the drugs, DMSO (Control and 50 mM KCl bars), or with 1 µM okadaic acid (Oka), 50 µM cantharidin (Canth), or 10 µM nifedipine (Nif) for 1 hr at 37 0C before being depolarized with KCl (50 mM KCl and +K bars). Results are reported as the mean from triplicate determinations ± SD. Single factor ANOVA was used to determine significant differences: P-values between KCl and Oka+K, Canth+K, and Nif+K are: 0.00043, 9.2xe-5, and 0.0056, respectively.
