Supplemental Figure. Each of the phosphorylation sites of the phosphoprotein DARPP-32 is shown with its respective phosphorylating enzyme. DARPP-32 is phosphorylated at Thr34 by PKA, at Thr75 by Cdk5, at Ser97 by CK2, and at Ser130 by CK1. P-Thr34 is preferentially dephosphorylated by PP-2B; P-Thr75 is preferentially dephosphorylated by PP-2A; P-Ser130 is preferentially dephosphorylated by PP-2C; the phosphatase for P-Ser97 is not yet characterized. Phosphorylation of Ser97 of DARPP-32 by CK2 increases the rate of phosphorylation of Thr34 by PKA; phosphorylation of Ser130 of DARPP-32 by CK1 decreases the rate of dephosphorylation of P-Thr34 by PP-2B. Phosphorylation at Thr75 converts DARPP-32 into an inhibitor of PKA, reducing its ability to phosphorylate DARPP-32 and other substrates (blue arrows indicate positive effect; red arrows indicate negative effect).
