Radial Migration of Superficial Layer Cortical Neurons Controlled by Novel Ig Cell Adhesion Molecule MDGA1

  • supplemental material - Supplemental Figure 1. cDNA sequence data of mouse MDGA1 Mouse MDGA1 cDNA was isolated from a cDNA library made from the basilar pons of C57BL/6 mice at P7. Basilar pons is a highly enriched expression domain of MDGA1 (Litwack et al., 2004). Total mRNA was isolated, reverse transcribed with oligo dT18 to generate a cDNA library, and RT-PCR was done with the primers designed based on rat MDGA1 cDNA sequences (Litwack et al., 2004) or partial mouse MDGA1 EST sequence in Genebank. A single full length mouse MDGA1 cDNA sequence was isolated and confirmed.
  • supplemental material - Supplemental Figure 2 . Transfected cells marked by GFP have a pyramidal morphology and express a neuronal marker, TuJ1. A. Morphology of cortical cells co-transfected with GFP and M6 RNAi vectors. Tuj1 mouse monoclonal antibody (1:500, Covance, CA USA) is visualized by the Alexa 568 goat anti-mouse antibody (Molecular Probes, OR, USA). The arrow points to a GFP labeled cell with a radially oriented bipolar morphology, positioned in superficial CP and positive for Tuj1. Scale bars = 10 μm.
  • supplemental material - Supplemental Figure 3. Transfections of M1 RNAi that diminishes MDGA1 expression preferentially affects superficial versus deep layer cortical neurons. Distribution of deep layer cortical neurons co-transfected with the GFP and E1 empty vectors (n=2) (A) or GFP and M1 RNAi vectors (n=2) (B). Transfections were done at E12.5; brains were fixed seven days later, late on P0, and coronally sectioned. The distributions of the GFP positive cells are similar between the two types of transfections and appropriate for the stage of transfection which should predominantly label deep player neurons. Abbreviations are as in Figure 1. Scale bar is 100 μm.
