Figure 1S: Chondroitinase ABC infused via gelfoam promotes mechanosensory axon regeneration into the spinal cord lesion cavity. A) Chondroitinase ABC treatment: Sensory axons (CTB-labeled, red fluorescence) grow into and past the lesion cavity as demarcated by GFAP-immunoreactivity. B) Control penicillinase treatment: Labeled axons stop caudal to the lesion cavity. C and D) To determine whether the applied chondroitinase ABC entered the injury site and degraded GAG chains, lesioned tissue was stained with anti-chondroitin-4-sulfate (ΔDi-4s) antibodies. C) Immunoreactivity for ΔDi-4s was found throughout the border of the lesion cavity 7-days after injury and treatment with chondroitinase ABC treatment. D) No anti-ΔDi-4s immunoreactivity was seen after penicillinase control infusion. E and F) Neurolucida reconstructions, see Materials and Methods, of sagital sections through the dorsal columns show the extent of fiber growth in a chondroitinase ABC and a penicillinase treated animal. G) To compare the magnitude of regeneration between neutralizing anti-NG2 monoclonal antibody and chondroitinase ABC treatments at 7-days post-lesion, axon density measurements were performed as described in Materials and Methods. No statistically significant differences were found at any distances from the lesion center between anti-NG2 (n=7) and chondroitinase treatment (n=6) (p≥0.08-1.0, one-way ANOVA with Bonferonni post hoc tests).
