A, Overexpression of the ‘non-phosphomimetic’ mutant of Syt1 mutated in both caskII phosphorlytion site (T125A/T128A) in knockout cells (grey; n = 20) resulted in similar secretion as overexpression of wild-type Syt1 (black; n = 19). (For detailed explanation of the traces see Figure 1A.) B, Overexpression of the ‘phosphomimetic’ T125D/T128D mutant of Syt1 (grey; n = 19) also did not change the secretion comparing to overexpression of wild-type Syt1 (black; n = 19). C, D, Kinetic parameters obtained from the triple exponential fit were normalized to the corresponding wild-type Syt1-overexpressing cells and displayed as mean±SEM. No change in the time constants (C), the amplitudes (D) of the two burst components, or in the sustained rate (D) was observed by the mutation.
