  • supplemental material - Supplemental Figure S1. Body weight gain of HPC-1/syntaxin 1A KO mice. Male offspring from HT mice intercrossing were weighed every week from Week 3 to Week 15. Lack of HPC-1/syntaxin 1A expression did not affect growth. (WT: n = 20; HT: n = 20; KO: n = 19) Data represent the mean + SD.
  • supplemental material - Supplemental Figure S2. Western blot analysis of HPC-1/syntaxin 1A and syntaxin 1B in hippocampus. Hippocampal tissue was obtained from twelve weeks old mice. 5 μg of the protein homogenate was used for Western blot analysis with anti-syntaxin 1 polyclonal antibody which equally reacted to syntaxin 1A and 1B. Immunoreactive bands were quantified using NIH Image. The content of HPC-1/syntaxin 1A and syntaxin 1B was nearly equal in hippocampus of WT mice (HPC-1/syntaxin 1A : syntaxin 1B = 1 : 0.86). The content of syntaxin 1B in hippocampus was not different among genotypes. (WT : HT : KO = 1+0.28 : 1.12+0.27 : 1.16+0.27, n=5). Data represent the mean + SD.

Left: Representative traces of spontaneous miniature excitatory postsynaptic currents (mEPSCs) at day in vitro at DIV 14 – 15. right: Normalized average trace of mEPSCs. The waveform of these mEPSCs did not different between KO and WT. Time constant of decay were 3.95 ms in WT and 3.79 ms in KO neurons.
  • supplemental material - Supplemental Figure S4. Schematic protocol of extinction training to cued fear memory. Each mouse received CS–UCS pairings five times in a conditioning chamber. Approximately 24 h after pairings, each mouse received cued fear memory extinction trials in a novel test chamber. The extinction trials consisted of CS for 57 s five times per day without USC in a novel test chamber. The freezing responses to CS were scored. The extinction trials were repeated for 4 days.
