Supplementary figure 1. The morphology of presynaptic dense projections following high-pressure freezing and freeze-substitution. Under these fixation conditions, the presynaptic density (black arrow) can be resolved into substructures, that is, electron dense projections that are ~30 nm in diameter. B and D, are high-zoom images of the outlined area in (A) and (C), respectively, which exemplifies the circular nature of the dense projections. (C), (E) and (F) are cross sections through the same neuromuscular junction. Bar equals 200 nm in (A), 100 nm in (C), (E) and (F), and 30 nm in (B) and (D).

Supplementary figure 2. A-D Representative micrographs for wild-type, unc-13(s69),unc-10(md1117) and unc-13(s69);unc-10(md117) neuromuscular junctions, respectively, following high-pressure freezing and freeze-substitution. Black arrows indicate dense projections; white arrows indicate examples of plasma membrane-contacting synaptic vesicles. Scale bar is 200 nm.
