  • supplemental material - Supplemental figure 1: SCH79797 blocks the calcium elevation produced by PAR-1 agonists (thrombin and TFLLR) but not by the PAR-2 agonist, trypsin. The y-axis shows the elevation of fluorescence of the calcium indicator, fluo-3, while the y-axis represents time of assay. The sharp spike present in the traces denotes the time of addition of the reagents.
  • supplemental material - Supplemental figure 2: Brain damage and the number of dying neurons are equivalent between groups whether blood from wildtype mice is injected into MMP-9 null brain, or whether blood from MMP-9 null animals is administered into wildtype brain. Each bar represents the mean ± SD of 5 animals.
  • supplemental material - Supplemental figure 3. Summary of results. Thrombin and MMP-9 have individual neurotoxic activity that is blocked by PAR-1 antagonist and MMP-9 inhibitor, respectively. Thrombin also acts through MMP-9 activation. Together, thrombin and MMP-9 interacts to increase the extent of neurotoxicity.
