  • supplemental material - Supplemental Figure 1. Diagram summarizing the injection protocol of the various 480 fragments. The experience was performed on 29 animals. One week after injection, 2 rats were analyzed to control the expression of huntingtin. 12 weeks after injection, the lesion was evaluated in one animal of each group. As the lesion was not significant at this time point, the final evaluation of all the animals was performed at 24 weeks after injection. The number of glutamines (17Q or 68Q) and the mutation of the serine 421 for each injected fragment of huntingtin are indicated on each hemisphere on the rat brain sections. n indicates the number of animals per condition.
  • supplemental material - Supplemental Figure 2. Rat striata injected with 480-17Q (wild-type), 480-68Q (polyQ), 480-68Q-S421A or 480-68Q-S421D lentiviruses were analyzed 24 weeks after injection by immunohistochemistry using anti-DARPP-32 and anti-phospho-S473-Akt (9277, Cell Signaling Technology) antibodies. Although a down-regulation of DARPP-32 is observed in the polyQ-huntingtin infected region, no signs of neuronal loss were observed as indicated by the presence of neurons immunopositive for the phosphorylated form of Akt.
