A, Fluorescence image of Munc13-1-EYFP puncta in cultured neurons isolated from the cerebral cortex of Munc13-1-EYFP mice and grown in vitro for 27 days. B, Same region as in (A) 14 hours after addition of MG132. To avoid the confounding effects of photobleaching, no images were obtained of this region between these two time points. Scale bar, 10 μm. C, Mean Munc13-1-EYFP puncta fluorescence after 12-14 hours, normalized to mean puncta fluorescence at time t=0. Mean and standard deviation for 15 fields of view recorded in a similar manner in 3 separate experiments (total of 6221 puncta at time 0 and 4957 puncta at time 12 to 14 hours).
