  • supplemental material - Supplemental Figure 1. Specificity of the TRPC7 antibody. Western blots of protein extracts from HEK 293 cells probed with α-TRPC7 (A) or α-TRPC7 pre-absorbed with antigen (B). Lanes represent (1) untransfected HEK293 cells, (2) HEK293 cells transfected with vector alone and (3) HEK293 cells transfected with vector + TRPC7 cDNA (marker indicates 100 kDa). The TRPC7 antibody recognised a 100 kDa protein in the HEK293 cells expressing the TRPC7 vector. HEK293 cells transfected with an epitope tagged TRPC7 vector (green) were treated with no antibody (control; C), α-TRPC7 (D) and α-TRPC7 pre-absorbed with antigen (E). TRPC7 expression (red) was observed following application of the TRPC7 antibody alone. Cells transfected with epitope tagged (green) TRPC1 (F), TRPC3 (G) and TRPC6 (H) were probed with α-TRPC7. No cross reactivity was observed. Scale bars represent 10 μm.
