Postnatal Generation of Neurons in the Ventrobasal Nucleus of the Rat Thalamus
J. Neurosci. Mooney and Miller 27: 5023

Supplemental Data

Files in this Data Supplement:

  • supplemental material - Supplemental Figure 1. Examples of NeuN-BrdU-co-localization. A. The merged, epifluorescence illuminated image shows multiple instances of co-localization of NeuN and BrdU (yellow arrows) in the VB. A cluster of cells was identified (box) that is the focus of images B - E. E. The merged image shows a newly generated neuron positive for all three labels (white arrow). Another cell was positive for BrdU and Hoescht and was NeuN-negative (purple arrow). Presumably it was a non-neuronal cell. One profile was NeuN-positive and Hoescht-negative (green arrow). This is likely the result of sectioning through the NeuN-positive perikaryon. F - H. Confocal images, with an optical section thickness of 1.0 ?m, show co-localization of cytoplasmic NeuN and nuclear BrdU. Scale bars are 50 ?m (A) and 10 ?m (B-H).
  • supplemental material - Supplemental Figure 2. HuC/D immunolabeling. Optical images were taken at different depths through the VB. The merged images (C and F) are also shown in Figure 7. A and D. HuC/D immunolabeling. B and E. BrdU positivity. Scale bars are 10 ?m.
  • supplemental material - Supplemental Figure 3. Co-localization of BrdU and a dextran injected into cortex. A. Dextran-positive cells were localized throughout the VB (green arrows). Inset shows the injection track through cortex. B. BrdU-positive cells (red arrows) were also found in the VB. C. Some dextran-positive cells in VB co-expressed BrdU (yellow arrows). Other cells expressed either the dextran (green arrows) or BrdU (red arrows). Scale bars are 50 ?m.
  • supplemental material - Supplemental Figure 4. Phenotype of non-thalamic cells. Animals injected with BrdU on P6 were euthanized on P12. A. Cells in cortex did not show co-localization of BrdU (green arrows) and NeuN (red arrows). B. Some cortical cells were double-labeled with BrdU and GFAP (yellow arrows). C - E. Some hippocampal cells co-localized BrdU and NeuN on P21 (yellow arrows). Scale bars are 50 ?m.
