Direct Astrocytic Contacts Regulate Local Maturation of Dendritic Spines
J. Neurosci. Nishida and Okabe 27: 331

Supplemental Data

Files in this Data Supplement:

  • supplemental material - Supplemental Figure 1 Astrocytic exclusive boundary and the vertical distribution of contact sites between dendrites and astrocytic processes in hippocampal slice cultures. a, Adjacent GFP-positive astrocytes show exclusive boundaries (broken lines) in a slice culture (5 DIV). Six image stacks (z-spacing 0.75 μm) were shown from the top to the bottom. b, The vertical distribution of astrocytes in a slice culture (5 DIV). A cryosection reveales few overlaps of GFAP-positive protrusions belonging to different cells. c, A cryosection cut vertically to the slice surface showing the contact site (arrow) between dendrites (red; labeled with rhodamine-dextran) and GFP-positive astrocytic processes (green) in the middle layer of the slice culture. In b and c, (A) and (B) indicate the surface and the bottom of the slice culture, respectively. Scale bars, a, 20 μm; b, 50 μm.
  • supplemental material - Supplemental Figure 2 Negligible effects of mutant Rac1 on the morphology of dendritic protrusions in neurons with a lower level of GFP expression. a, Morphology of dendrites visualized by DiI application after infection with bicistronic recombinant adenoviruses for simultaneous expression of mutant Rac1 and GFP. Although dendritic protrusions in cells with intense GFP signal, which indicates concomitant higher expression of mutant Rac1, show elongated morphology, cells with lower GFP signal have dendritic protrusions indistinguishable from control. Scale bar, 5 μm. b, Negligible effects of mutant Rac1 on the length of dendritic protrusions in cells expressing different levels of GFP and mutant Rac1. ANOVA with post hoc multiple comparisons by Tukey’s test indicated no statistical difference among groups (p > 0.05).
