Sup Figure 1. Kynurenate increases the potency of 5-HT mediated inhibition indicative of a reduction in the peak glutamate cleft concentration.
A, 300nM 5-HT (dark gray) reduced the EPSC to 46% of control (black). 200µM Kyn (gray, bottom) reduced the EPSC to 48% of control. Application of 300nM 5-HT with Kyn (light gray, bottom) reduced the EPSC to 14% of the Kyn EPSC (gray).
B, EPSCs for all 4 conditions. Subtraction of the electrical component of the EPSCs reveals isolated AMPA-receptor responses (left). EPSCs recorded in 300nM 5-HT with Kyn (light gray) and 200µM Kyn (gray) are scaled (dashed lines) to control (black, right). Kyn increased the potency of 300nM 5-HT (light gray versus gray traces).
C, Bar graph depicting the inhibitory potency of 5-HT in control versus the potency of 5-HT in the presence of Kyn (200µM) for 4 different experiments. Averaging the responses revealed that 300nM 5-HT alone inhibited the chemical component of the EPSC to 60.7 ± 6.5% of control (solid gray bar). However, in 200?M Kyn the same dose of 5-HT (300nM) inhibited synaptic transmission to 35.2 ± 10.6% of the response in Kyn alone (solid light gray bar, significantly different, p<0.05, n=4). EPSC responses in 5-HT (solid gray bar) are normalized to control (open black bar), and EPSC responses in 5-HT and Kyn (solid light gray bar) are normalized to the response in Kyn alone (open gray bar, * denotes significant difference for 5-HT and Kyn vs. 5-HT alone, p<0.05).

Supplemental figure 2. 5-HT does not alter the electrical component of the EPSC. The peak amplitude of the electrical component of EPSCs is plotted versus time. The electrical component varies slightly, but the amplitude is not altered by 5-HT. Red line represents a linear fit of the data. Arrow denotes bath application of 300nM 5-HT.
