Variations in Promoter Activity Reveal a Differential Expression and Physiology of Glutamate Transporters by Glia in the Developing and Mature CNS
J. Neurosci. Regan et al. 27: 6607

Supplemental Data

Files in this Data Supplement:

  • supplemental material - Supplemental Figure 1. Differential GLAST and GLT-1 promoter activation in cerebral cortex. A) Analysis of cortex from P24 GLAST-DsRed/GLT-1-eGFP double transgenic mice reveals prominent GLT-1 promoter activation in astrocytes along with numerous (red arrows), but not all (yellow arrows), neurons exhibiting nuclear expression of the GLAST promoter reporter DsRed, as indicated by neuronal marker NeuN (blue). The presence of this GLAST promoter reporter neuronal nuclear fluorescence is initially restricted to lower cortical layers and periventricular regions, and is localized to mid-cortical layers with aging. The GLAST neuronal expression decreases with aging and is absent by P50. Scale bar=50 µM. B and C) High power confocal images of cortex from the mid-cortical layers of P24 GLAST-DsRed/GLT-1-eGFP double transgenic mice also demonstrate differential GLAST and GLT-1 promoter activation. * marks cortical neurons. Confocal image thickness, B=1µM; C=10 µM (collapsed stack). Scale bar = 20 µM.
