Supplementary Figure 1.
(A) Sequences used for the construction of shRNA vectors (shNR1a-h). Target sequences are in bold.
(B) Screen of shNR1 vectors for activation of an interferon-like response in cultured dissociated hippocampal neurons (Alvarez et al., 2006). Luciferase was transcribed from an interferon-inducable promoter (mx-2) and its activity was measured by luminescence (2 DPT, n=3 experiments). Values are ratios between inducible- and constitutively-expressed luciferases expressed as percentage of the ratios in control neurons expressing GFP alone. * indicates P < 0.01, ANOVA.

Supplementary Figure 2.
The number of branch points in the apical and basal dendrites of control (GFP transfected neurons) and shNR1 expressing neurons in organotypic slice culture (n= 7-15 cells, ANOVA). No statistical difference was found across conditions consistent with no effect of NMDAR loss on dendrite complexity.
