S2. Biochemical analyses of TDP-43 in sporadic and familial (Gly333ValfsX28 PGRN mutation) FTLD-U (Gass et al., 2006) AD and vascular dementia (VaD) brains. Immunoblots of Triton-X soluble (Sol) and Triton-X insoluble (Ins) fractions from temporal cortex of cases and controls with rabbit anti-TDP-43 antibody shows a similar biochemical profile of TDP-43 to that observed in progranulin knockdown experiments in cultured cells (Figure 1B and 2A).

S3. Histone immunocytochemistry demonstrates mostly nuclear staining in H4 cell treated with 1 µM staurosporine, demonstrating that the TDP-43 subcellular redistribution is unlikely to be related to nonspecific leakage of DNA binding proteins from nucleus to cytoplasm induced by apoptosis.
