To illustrate bilateral retinal innervation of the SCN, retinal fibers were labeled after monocular injection of HRP into the vitreous body of the eye. HRP-labeled optic fibers are visible in this dark-field image in the opticnerve (CN II) of the injected eye, crossing in the optic chiasm, and termination bilaterally in the SCN just posterior to the optic chiasm. Whole-cell patch-clamp recordings of evoked EPSCs in SCN neurons after optic nerve stimulation were conducted to demonstrate 5-HT1B receptor-mediated presynaptic inhibition of retinohypothalamic tract neurotransmission to the SCN.
