Cytochrome c activates the adapter molecule APAF-1, resulting in the formation of the apoptosome, a multiprotein complex in which the initiator caspase-9 is activated (12) for processing of caspase-3 and amplification of the caspase cascade. Upon TRAIL-R ligation, MOMP is induced by caspase-8-mediated cleavage and activation of BH3-interacting domain death agonist (Bid), a proapoptotic protein of the B cell lymphoma 2 (Bcl-2) family (13–15). The proteins of the Bcl-2 family are key regulators of MOMP and show homology in at least one of four Bcl-2 homology (BH1–4) domains.
