LNA-modified ASOs are known to have higher affinity for their target compared with earlier generations of chemically modified ASOs.33 Here we found that ASO-001933 given at a dose of 8 mg × 2 (16 mg total) produced up to 80% knockdown of both tau transcript and protein in cortex and hippocampus.
