Functions and distribution of calpain-calpastatin system components in brain during mammal ontogeny
2023, Biochimica et Biophysica Acta - General SubjectsCalpain inhibition reduces NMDA receptor rundown in rat substantia nigra dopamine neurons
2018, NeuropharmacologyCitation Excerpt :Calpain expression is also isoform-dependent, with calpain-1 being expressed in neuronal somata and calpain-2 expressed in axonal processes and glial cells (Siman et al., 1985). Furthermore, higher levels of calpain are found in hindbrain and midbrain structures compared with forebrain structures in rats (Simonson et al., 1985). Calpain is present in SNc-DA neurons in the adult rat (Siman et al., 1985), although in human post-mortem brains, calpain-2 was detected only in the substantia nigra of Parkinsonian (PD) patients, not in control subjects (Mouatt-Prigent et al., 1996), raising the possibility that calpain-2 might be upregulated in PD.
Defects in the CAPN1 Gene Result in Alterations in Cerebellar Development and Cerebellar Ataxia in Mice and Humans
2016, Cell ReportsCitation Excerpt :Immunohistochemistry study revealed that the major calpain isoform expressed in cerebellar neurons is calpain-1 (Hamakubo et al., 1986). Calpain-1 activity in cerebellum during prenatal and early postnatal period is high, as compared to that in adulthood (Simonson et al., 1985), suggesting a potential role for calpain-1 in cerebellar development. Interestingly, a CAPN1 missense mutation in the Parson Russell Terrier dog breed has been associated with spinocerebellar ataxia (Forman et al., 2013).
Calpain-mediated regulation of stargazin in adult rat brain
2011, NeuroscienceCitation Excerpt :In the cerebellum, stargazin immunoreactivity was higher in the molecular layer than in the granule layer, and calcium-induced decrease in stargazin levels was relatively similar in the molecular and granule layers. Several studies have shown that calpain was present both in Purkinje cells and granule cells, although it is difficult to determine the relative distribution of calpain between these two neuronal populations (Simonson et al., 1985; Hamakubo et al., 1986; Li et al., 1996; Shields et al., 1998). In addition, we did not attempt to determine whether calpain-1 or calpain-2 was involved in calcium-induced changes in stargazin levels, since, under our experimental conditions, both types of calpain should be activated.
