2023, NeuroscienceCitation Excerpt :Many more subtypes of Semaphorins have been identified since then and characterized as axon guidance cues throughout the peripheral and the central nervous systems (Pasterkamp, 2012; Jongbloets and Pasterkamp, 2014; Limoni, 2021). An extremely helpful in vitro assay for the analysis of diffusible axon guidance cues, such as Semaphorin3A, was the 3D collagen gel assay, originally described by Ebendal and Jacobson (Ebendal and Jacobson, 1977) (Fig. 3(A,B)). This assay has also been used to demonstrate positive/attractive activity on growing neurites (e.g. (Lumsden and Davies, 1983)).

2009, NeuronCitation Excerpt :To investigate the molecular mechanism of Shh in axon guidance, we developed an axon guidance assay that functioned over short time scales (1–2 hr) in commissural neurons and could acutely inhibit transcription or other signaling pathways without affecting cell fate. Existing collagen gel explant assays (e.g., Ebendal and Jacobson, 1977; Serafini et al., 1994) were not suitable since they require 1–2 days to perform, and many of them typically measure preferential growth, rather than direct turning, toward a guidance cue. The pipette turning assay (Gundersen and Barrett, 1979; Lohof et al., 1992) does measure turning in response to a local gradient of chemoattractant in ∼1 hr.
