Protein traffic between distinct plasma membrane domains: Isolation and characterization of vesicular carriers involved in transcytosis

Abstract

We have Isolated a population of vesicular carriers involved in the transport (transcytosis) of proteins from the basolateral to the apical plasma membrane of hepatocytes. The obtained fraction was enriched in compartments containing known transcytosed proteins and depleted in elements of the secretory pathway, Golgi elements, basolateral plasma membrane, as well as early endosomal components. The fraction was analyzed by biochemical and Immunological procedures. Antibodies raised against the proteins in the fraction recognized a single 108K antigen. Based on its subcellular distribution, the 108K antigen may represent a novel marker for transcytotic vesicular carriers.

      Cholesterol content in each fraction was determined enzymatically in a microassay using a diagnostic kit from Roche Molecular Biochemicals (35) and procedures reported by our laboratory (36-38). A marker enzyme of the Golgi complex is galactosyltransferase, and activity of this enzyme was measured using procedures reported previously (32, 39). The incubation mixture contained 7 mg/ml ovalbumin, 2 mm ATP, 200 mm MgCl2, 0.27 Triton X-100, and 50 mm Tris-HCl (pH 6.8).

      The method of Lowry et al. (26) was used for protein measurements with bovine serum albumin as the standard. The activities of marker enzymes were determined according to the following protocols: γ-glutamyl transpeptidase (27), alkaline phosphatase (28), Na,K-ATPase (29), and galactosyltransferase (30). The quality of the membrane preparations was determined by measuring the enrichment of marker enzymes (Table I).
