Hair Cell Mechanotransduction Regulates Spontaneous Activity and Spiral Ganglion Subtype Specification in the Auditory System
2018, CellCitation Excerpt :Labeled sections were imaged using a Zeiss 700 Confocal Laser Microscope System. For primary antibodies, we used goat α-CALB2 (CG1, Swant, 1:2500), mouse α-CALB2 (MAB1568, Millipore, 1:1000), mouse α-TuJ1 (801201, Biolegend, 1:400), rabbit α-TuJ1 (SAB4500088, Sigma, 1:200), mouse α-POU4F1 (MAB1585, Millipore, 1:20), rabbit α-CALB1 (13176, New England Biolabs, 1:50), rabbit α-Tyrosine Hydroxylase (657012, Millipore, 1:1000) and rabbit α-p75 NGFR (Weskamp and Reichardt, 1991, 1:800). For secondary antibodies, we used goat α-rabbit (A11008, Alexa 488 conjugated; A11011, Alexa 568 conjugated, Thermo Fisher), goat α-mouse (A11001.
Somatodendritic Expression of JAM2 Inhibits Oligodendrocyte Myelination
2016, NeuronCitation Excerpt :Spinal cords were chopped with a sterile scalpel blade, and the tissue was incubated in 0.05% trypsin-EDTA (Life Technologies) for 15 min at 37°C. The tissue was dissociated into a single-cell suspension and incubated in a series of immunopanning dishes: two negative-selection dishes coated with monoclonal hybridoma antibodies rat neural antigen-2 (Ran-2, ATCC) and galactocerebroside (Gal-C; Ranscht et al., 1982) and one positive-selection plate against p75 neurotrophin receptor (P75NTR; rat cells, MC192 hybridoma; mouse cells, rabbit anti-P75NTR function blocking antibody “REX,” a generous gift of L.F. Reichardt; Weskamp and Reichardt, 1991). Adherent cells were released from the final immunopanning plate with a brief application of 0.05% trypsin-EDTA, and 200,000–300,000 cells were seeded on 25 mm round coverslips prepared with 200 μL of 1:25 Matrigel (Corning, 356230) in DMEM (Life Technologies).
