2012, NeuroscienceCitation Excerpt :Although most high-voltage activated (HVA) as well as some low-voltage activated (LVA) Ca2+ channels contribute to spike-triggered Ca2+ entry in the soma and basal dendritic region (Christie et al., 1995), Cav2.3 R-type and T-type VGCCs predominantly underlie spike-triggered Ca2+ entry in apical dendrites (Christie et al., 1995; Magee and Johnston, 1995; Sabatini and Svoboda, 2000; Yasuda et al., 2003). Paradoxically, muscarinic stimulation increases [Ca2+]i in dendrites and spines following depolarization or synaptic stimulation, and it was hypothesized that K+ conductance blockade and/or IP3-mediated Ca2+ release from intracellular stores are related to this [Ca2+]i accumulation (Muller and Connor, 1991, 1992; Tsubokawa and Ross, 1997; Beier and Barish, 2000). Although muscarinic stimulation is known to suppress K+ currents, for example, IM, thus mediating enhanced neuronal excitability (Brown and Adams, 1980; Halliwell and Adams, 1982), in vitro studies by Tai et al. (2006) showed that the enhancement of R-type Ca2+ current is not related to K+ channel depression and hippocampal K+ conductance was not affected by M1 receptor activation (Fisahn et al., 2002).

The initial [Ca2+]i increase activates high threshold plasma membrane channels which then mediate further Ca2+ release (“Ca2+-dependent Ca2+-release”) [21,33]. The depressant effects of dantrolene and thapsigargin in our experiments suggest that the epileptogenic effects of CCh or endogenous ACh (eserine) may be partially mediated by a muscarinic receptor-induced increase in [Ca2+]i.
