In both mouse lines, GFP+ axonal boutons exhibit a high degree of overlap with VGluT2 immunostaining (Figure 1E).

2022, NeuroscienceCitation Excerpt :The slides were washed in PBS followed by blocking and permeabilization for 1 hour in blocking buffer (PBS, 0.5% Triton X-100, 5.5% donkey serum, 5.5% goat serum, pH ∼ 7.4). The slides were then incubated in primary antibodies diluted in blocking buffer (1:250 guinea pig-anti-vGlut2, Millipore AB255; 1:500 rabbit-anti-Iba1, Wako 019-19741) overnight – vGlut2 is a selective marker for retinogeniculate terminals in the dLGN (Fujiyama et al., 2003; Land et al., 2004; Yoshida et al., 2009; Koch et al., 2011; Seabrook et al., 2013), while Iba1 was used to label microglia. Following incubation with primary antibodies, the slices were washed in PBS, blocked for 1 hour and incubated with secondary antibodies diluted in blocking buffer (1:200, goat-anti-guinea pig IgG, Alexa Fluor 488, Invitrogen A11073; 1:200 donkey-anti-rabbit IgG, Alexa Fluor 568, Invitrogen A10042), washed and coverslipped with Vectashield Hardset mounting medium and stored at 4 °C until imaged.
CD47 Protects Synapses from Excess Microglia-Mediated Pruning during Development
2018, NeuronCitation Excerpt :Immunohistochemistry (IHC) for CD47 revealed an enrichment in the dLGN during peak pruning at postnatal day (P) 5 (Figures 1A and S1A), when microglia are highly phagocytic and have been shown to engulf RGC inputs (Schafer et al., 2012). We confirmed CD47 localization to this region by costaining for vesicular glutamate transporter 2 (Vglut2), a presynaptic marker specific for RGC terminals in the dLGN (Figure 1B) (Land et al., 2004). At P10 (Figure 1A), CD47 remained elevated in the dLGN compared to other brain regions, but by P30 (Figure 1A), when eye-specific segregation is largely complete, it was uniformly localized throughout the neuropil in all brain regions examined, as expected for a ubiquitous protective signal.
Refinement of the Retinogeniculate Synapse by Bouton Clustering
2014, NeuronCitation Excerpt :Thus, an increase in bouton size may correspond to the addition of synapses within each bouton as connections are strengthened over development. In order to determine whether structural boutons contained presynaptic elements, we quantified the degree of colocalization of boutons with vesicular glutamate transporter 2 (VGLUT2), a presynaptic marker for RGC, but not cortical, inputs in the LGN (Land et al., 2004) (Figures 2D–2I). Colocalization was high at both immature and mature axons and did not significantly differ with age (88.2% ± 4.1% at P8 versus 92.5% ± 6.5% at P20–P30, SD, p = 0.09, Student’s t test, n = 10, statistical power = 0.8; Supplemental Experimental Procedures; Figure S2G).
