This super-resolution Structured Illumination Microscopy image illustrates enlarged, permeabilized lysosomes of auditory outer hair cells lacking lysosomal channel mucolipins 1 and 3. In the co-absence of these channels, lysosomes (red) become “leaky”, allowing cytosolic galectin 3 (green) to enter and bind to luminal glycocalyx. The permeabilized lysosomes also let toxic hydrolases into the cytoplasm which causes hair cell death and, therefore, hearing loss. (pages 3177–3189).
