2021, Neurobiology of Learning and MemoryCitation Excerpt :Moreover, when the paired-pulse index was determined before and after HFS application, we found a significant decrease at the inter-pulse interval of 80 ms, in the autistic-like group compared to control rats. It has been demonstrated that at short inter-pulse intervals of less than 50 ms, the decrease in response to the second stimulus is attributed to GABAA receptor-mediated inhibition, while slow synaptic inhibition mediated by GABAB receptor dominates at intervals between 100 and 125 ms activation of presynaptic (Steffensen & Henriksen, 1991). Thus, in the present study, it is difficult to attribute the decrease in paired-pulse ratio at 80 ms inter-pulse interval directly to the GABA receptors function in the prenatal VPA-exposed rats.

Previous study in our laboratory (Leung et al., 2008) showed that paired-pulse inhibition of the proximal apical dendritic sinks evoked by CA3 homosynaptic paired-pulse stimulation was blocked by GABAA receptor antagonist picrotoxin, suggesting a postsynaptic GABAA receptor-mediated inhibition (Steffensen and Henriksen, 1991), possibly by basket cells (Freund and Buzsaki, 1996; Leung et al., 2008). However, the lack of CA1 PS inhibition suggests a layer-specific re-distribution of inhibition, but not a decrease in total inhibition of spike firing.

Paired-pulse facilitation of population spike develops at longer inter-pulse intervals and this effect can be promoted by an augmentation in glutamatergic and a reduction in GABAergic transmission, respectively (Bordi et al., 1997; Brucato, Morrisett, Wilson, & Swartzwelder, 1992; Canning & Leung, 2000; Joy & Albertson, 1993; Steffensen & Henriksen, 1991). Our results suggest that CB1 might suppress inhibitory output of GABAergic neurons onto granule cells in the dentate gyrus.
