In vivo extracellular recordings have shown that MPOA contains cells with diverse response patterns during sexual behaviors (Horio et al., 1986; Shimura et al., 1994): some MPOA cells are excited during pursuit of females and mounting initiation but inhibited during intromission, while others are activated only during advanced stages of sexual behaviors such as intromission and ejaculation. In vivo calcium imaging of estrogen receptor alpha (Esr1)-expressing cells in the MPOA (MPOAEsr1+) showed female-preferred responses and ramping activity during mounting (Karigo et al., 2021; Wei et al., 2018).

During the various phases of sexual behavior, a large increase in c-Fos levels is observed in rats, together with increased electrical activity in distinct mPOA group of cells (Shimura et al., 1994). This indicates that there are functional subdivisions in the mPOA contributing to sexual motivation and copulatory performances (Baum and Everitt, 1992; Coolen et al., 1998; Hull et al., 2002; Paredes, 2003; Pfaus and Heeb, 1997).
