TRPA1 activation mediates nociception behaviors in a mouse model of relapsing-remitting experimental autoimmune encephalomyelitis
2020, Experimental NeurologyCitation Excerpt :The EAE model with PLP is an RRMS model, but it is necessary to use old SJL mice because they are more susceptible to the development of demyelination lesions with this antigen. Another disadvantage is the absence of demyelination at dorsal root entry (Chalk et al., 1994; Rasmussen et al., 2007). Using MOG35–55 as antigen and complete Freund's adjuvant (CFA), it is possible to reproduce a progressive MS model; however, the animals show severe motor impairment that can impair the measurements of nociception (Khan et al., 2014; Olechowski et al., 2013).
Involvement of JAK/STAT signaling in the effect of cornel iridoid glycoside on experimental autoimmune encephalomyelitis amelioration in rats
2014, Journal of NeuroimmunologyCitation Excerpt :To assess the degree of inflammation in spinal cords, HE stained sections were counted under a light microscope at 20 × magnification. The extent of mononuclear cell infiltration was evaluated as follows: 0, absent; 1, very mild; 2, mild (2 perivascular cuffs per spinal cord section); 3, moderate (3 perivascular cuffs per spinal cord section); 4, severe (N3 perivascular cuffs per spinal cord section) (Chalk et al., 1994). For all immunostaining, the spinal cords were cut coronally in 20–40 μm serial sections using a Leica cryostat and kept free floating.
Epimedium flavonoids ameliorate experimental autoimmune encephalomyelitis in rats by modulating neuroinflammatory and neurotrophic responses
2012, NeuropharmacologyCitation Excerpt :To assess the degree of inflammation in the spinal cords, HE & LFB stained sections were counted under a light microscope at 20× magnifications. The extent of mononuclear cell infiltration was evaluated: 0, absent; 1, very mild; 2, mild (2 perivascular cuffs per spinal cord section); 3, moderate (3 perivascular cuffs per spinal cord section); 4, severe (N3 perivascular cuffs per spinal cord section) (Chalk et al., 1994). For all immunostaining, the sections were first rinsed with PBS containing 0.1% Triton-X (PBS-T), and then were immersed in a solution of 0.5% H2O2 for 30 min.
Myelin proteolipid protein: An effective autoantigen and target of autoimmunity in multiple sclerosis
2008, Journal of AutoimmunityCitation Excerpt :In EAE, histopathological analysis and neurophysiological studies have shown that PLP-induced lesions are restricted to the CNS, with no involvement of the PNS [8,12,15–17]. The absence of PNS involvement in PLP-induced EAE is in marked contrast to the major involvement of the spinal nerve roots in the PNS in MBP-induced EAE [16,17]. PLP has a long half-life and the turn-over of PLP is low.
