Stress Resilience is Associated with Hippocampal Synaptoprotection in the Female Rat Learned Helplessness Paradigm
2021, NeuroscienceCitation Excerpt :The goal of this study, however, is to prevent the development of helpless behavior via promoting stress resilience, which logically requires drug application prior to stress exposure. This approach obviously follows the setup of earlier learned helplessness studies that revealed the preventive effects of benzodiazepines and ovarian hormones (Drugan et al., 1984; De Pablo et al., 1991; Petty et al., 1992; Baka et al., 2017). Here we report that promoting stress resilience in learned helplessness with diazepam is associated with a synaptoprotective action in the hippocampus and with a muted stress response during escape testing.

Chronic stress results in increased 5-HT turn-over in the hippocampus and frontal cortex (Petty et al., 1992; Torres et al., 2002; Gamaro et al., 2003; Ahmad et al., 2010) and this can be prevented by chronic treatment with antidepressants (dos Santos et al., 2010). In contrast, MDMA-induced monoamine depletion is enhanced by chronic unpredictable stress (Johnson and Yamamoto, 2010) which also potentiates the 5-HT2A-mediated effects of methamphetamine (Doyle and Yamamoto, 2010).
