2019, Brain ResearchCitation Excerpt :At least some of these acidic hydrolases have been implicated in the pathogenesis of neuroHIV (Rivera et al., 2014; Cantres-Rosario et al., 2019). For neurons, endolysosomes are especially important because neurons are mainly long-lived post-mitotic cells that require endolysosomes in turning over cellular components and obsolete organelles (Nixon and Cataldo, 1995; Nixon and Cataldo, 2006). The acidic pH is also important for the fusing of autophagosomes with lysosomes to form autophagolysosomes; a process known as autophagy (Tooze and Dikic, 2016).
The Temporal Dynamics of Arc Expression Regulate Cognitive Flexibility
2018, NeuronCitation Excerpt :However, this does not appear to be involved in AMPAR endocytosis as mutation of this site in addition to K268 and K269 did not further alter AMPAR internalization (Figures S1B and S1C). An alternative interpretation is that the lysosome-dependent pathway regulates Arc degradation (Nixon and Cataldo, 1995). Recent findings suggest that Arc protein and mRNA undergo self-intercellular transfer by assembling into virus-like capsids.
Hippocampal plasticity during the progression of Alzheimer's disease
2015, NeuroscienceCitation Excerpt :The underlying pathogenic mechanisms that trigger these structural synaptic alterations in the hippocampus during MCI and the onset of AD are unclear, yet several lines of evidence revealed that an aberrant expression of endosomal-lysosomal proteins occurs within hippocampal neurons even before SP and NFT formation early in AD (Nixon et al., 1992; Cataldo et al., 1994, 1995; Nixon, 2005). Endosomal-lysosomal activity is an essential and dynamic organization of acidified cytoplasmic organelles which perform a variety of functions in neurons such as internalizing nutrients and neurotrophic factors, as well as degrading and recycling receptors (Nixon and Cataldo, 1995; Bishop, 2003). Acid hydrolases such as cathepsins are transported from the Golgi apparatus to late endosomes that contain intracellular material engulfed during autophagy or extracellular material derived from heterophagy (Gordon and Seglen, 1988; Dunn, 1990; Gordon et al., 1992).
