The Neuropeptide Galanin Is Required for Homeostatic Rebound Sleep following Increased Neuronal Activity
2019, NeuronCitation Excerpt :These data indicate that the homolog of at least one important sleep-active center of the mammalian brain, the Galn expressing VLPO, is also active during rebound sleep in zebrafish. Increased neuronal activity is often accompanied by changes in neuropeptide transcription rates via “stimulus-secretion-synthesis coupling” (MacArthur and Eiden, 1996), presumably as a mechanism to replenish neuropeptide stores after secretion. We therefore tested whether the increased Galn neuron activity during rebound sleep was accompanied by upregulated galn transcription.

This interpretation is consistent with stimulus-secretion-synthesis coupling that has been demonstrated in other neuropeptide systems (MacArthur and Eiden, 1996). Clearly, an expanded time course of EB effects on OT mRNA and protein expression will be necessary to further explore the timing of OT synthesis, transport, and release, as well as the implication of these findings on the timing of EB-mediated effects on feeding.

Evidence has been provided that PACAP up‐regulates its own expression in rat adrenomedullary cells, via a mechanism involving the activation of AC‐ and PLC‐dependent cascades and an increase in the intracellular Ca2+ concentration ([Ca2+]i) (MacArthur and Eiden, 1996). Likewise, in the presence of nerve growth factor (NGF), PACAP38 has been shown to enhance PACAP mRNA in PC12 cells, through the activation of a p38 mitogen‐activated protein kinase (MAPK) cascade (Sakai et al., 2004).
