We and others have shown that the PKC pathway is very important in mediating insulin secretion from β-cells in response to cholinergic stimulation (Jones et al., 1991; Persaud, 1997; Persaud and Jones, 1993, 1994, 1995; Persaud et al., 1989, 1991, 1992; Tian et al., 1996). Indeed, typical, novel and atypical PKCs are expressed by β-cells and they modulate insulin secretion following cholinergic activation (Ahmed et al., 2015; Fleming and Storz, 2017; Knutson and Hoenig, 1994; Tang and Sharp, 1998; Tian et al., 1996; Velazquez-Garcia et al., 2011; Warwar et al., 2006).

The involvement of various PKC isoforms in the phosphorylation of downstream protein substrates and in regulation of PLC generated intracellular Ca2+ ions has been reported to be important for insulin secretion (Tian et al., 1996; Zawalich and Zawalich, 2001; Zhang et al., 2004; Cheng et al., 2012). The hydrolysate used in our study induced insulin secretion by activating PLC-PKC which is evident from decreased response of insulin secretion in presence of the PLC inhibitor U-73122 and PKC inhibitor Go-6983 (Macmillan and McCarron, 2010; Young et al., 2005).

Within normal tissue, glucose is the main stimulator of insulin release and this is PKC-dependent as evidenced by usage of the pan-PKC inhibitor Ro 31-8220, which blocks the release of insulin [11].
