Studies in animals demonstrate a direct anatomical connection between the main sources of serotonin in the brain, the brainstem dorsal and median raphe nuclei, and forebrain limbic structures, such as the amygdala, that control anxiety and fear responses (Fanselow and LeDoux, 1999; Fernandez et al., 2016; LaBar et al., 1998; Parent et al., 1981). Exposure to conditioned stimuli in the FPS paradigm increases c-Fos expression, a marker of neuronal activation, in serotonergic neurons within the dorsal raphe nucleus as well as the amygdala (Spannuth et al., 2011).
